Electric blue is a fusion of 2 fluorescent/chromoproteins: amilCP and Electra2. The former contributes a deep blue purple shade in white light, while the former contributes it’s ultramarine fluorescent glow under UV. The plasmid is designed so that you can easily pop the protein out and transplant it into your own constructs and use it as a reporter. Or just use it for some beautiful agar art.The plasmid is based on pet9a, and as such uses kanamycin as the resistance marker, and is low copy. The DNA is provided as 10ug of freeze dried DNA and must be shipped express. Freeze and store at -20C on receipt. As we are rolling out our first DNA Sale there is up to a 2 month lead time. This plasmid is all about smells. Specifically, acetate esters. These are fruity smelling compounds that are made by combining various alcohols with acetate groups. This plasmid expresses ATF1 from Saccharomyces cerevisiae, which is an enzyme that will perform the esterification reaction at room temperature with the assistance of Acetyl-CoA, which the bacteria produce naturally. The enzyme is on a strong constitutive promoter so that not a lot of growth time is required to attain sufficient quantities of the enzyme for the reaction to happen. This is to limit the amount of indole that will build up, as E. coli naturally emit indole which has a fecale odour. For this to work, you simply transfect your E. coli strain of choice with the plasmid, and then innoculate 1-10ml of LB with the modified bacteria. Let it grow for an hour or two, then add a few microliters of your alcohol of choice to the tube. The classic is isoamyl alcohol as it will produce a distinct banana smell when combined with the modified bacteria. But feel free to experiment with other alcohols. Hexanol or octanol are good choices as well. The plasmid is based on pet9a, and as such uses kanamycin as the resistance marker, and is low copy. As we are rolling out our first DNA Sale there is up to a 2 month lead time. This plasmid is an implementation of a simple transistor circuit. There are two sections of the plasmid, the transistor part, and the reporter part. The transistor part is composed of the lac and tet repressors wired to control each other. The reporter part consists of 2 fluorescent proteins to indicate what state the system is in. When the plasmid is initially transformed into your E. coli strain of choice, it will settle randomly into one of the two states: red, and green. When streaked out onto plates containing lactose or IPTG, the switch will engage, and lock into the red state. This state should be maintained for the most part even if the bacteria are streaked out back on regular antibiotic plates. However if the bacteria are streaked onto plates containing anhydrotetracycline (chosen for it’s ability to trigger the tet repressor without having antibiotic activity), the system will switch into a green/yellow state. Again this state should be stable until lactose is given.The plasmid is based on pet9a, and as such uses kanamycin as the resistance marker, and is low copy. As we are rolling out our first DNA Sale there is up to a 2 month lead time. This plasmid is our take on a classic and is a demonstration of a transistor-like oscillation circuit. It uses 3 repressor proteins wired in a loop to create a continuous oscillation inside living bacteria, effectively forming a clock. There are 3 reporter proteins so you can see what state the clock is in at any moment. Two of the repressors are chemically triggerable, so you can shift what the oscillations do by streaking the bacteria onto plates that contain either lactose or anhydrotetracycline. This plasmid is what we’re calling our “base” clock speed. The various colonies on a plate of this construct will glow different colors and may even display a ring structure as the clock ticks in each colony.We are planning to release a faster clock that enhances the ring effect at a later date.The plasmid is based on pet9a, and as such uses kanamycin as the resistance marker, and is low copy.
